Abstract

Disclosed is a method for inducing a gardenia embryonic callus, relating to the technical field of plant tissue culture. The method comprises the steps of inoculating a gardenia explant into a culture medium for inducing the gardenia explant to generate an embryonic callus, performing dark culturing, and obtaining the gardenia embryonic callus. The culture medium comprises 0.1-0.3 mg/L 6-BA, 0.1-0.3 mg/L 2,4-D, and 12 mg/L gardenia fruit extract. The effects of different types of plant growth regulators and different test materials on the induction of gardenia embryonic callus were studied, the ratio and concentration of plant growth regulators were optimized, and the gardenia fruit extract was applied to the embryonic callus induction medium, thereby greatly improving the induction rate of gardenia embryonic callus.
