The second control unit generates a data packet comprising the position coordinates and sending the data packet to the first control unit. The second control unit also generates trigger signals for the photodetector, the trigger signals being configured to trigger the photodetector to capture images corresponding to each of the position coordinates of the movable mirror. The first control unit and the second control unit communicate over Ethernet such that synchronization between the steering of the movable mirror and the triggering of the photodetector is enabled.

The retaining mechanism is configured to retain a sample between the holding component of the retaining mechanism and the first surface area in a closed state of the retaining mechanism. The retaining mechanism is further configured to enter an opened state upon mechanical actuation of the retaining mechanism. The opened state allows insertion and removal of the sample. The retaining mechanism is further configured to revert to the closed state once the mechanical actuation is removed.

Abstract

A microtome system for cutting sections from a specimen includes a knife including a knife edge configured to cut a section from the specimen, a knife holder, a specimen holder, an illumination, a first actor, a detector, and a controller. The knife holder and specimen holder are configured to be relatively moveable in a cutting direction. The first actor is configured to cause a rotation of the knife holder or specimen holder about an axis. The illumination is configured to illuminate a gap between a front face of the specimen and the knife edge to generate a light gap. The detector is configured to detect a geometric feature of the light gap. The controller is configured to automatically align, or provide indications to manually align, the knife edge with the front face, by controlling the first actor depending on the detected geometric feature.

Abstract

A method is proposed for generating a series of ultra-thin sections of a microscopic sample (10), wherein the sections (11) are detached from the sample (10) using an ultramicrotome (100) and wherein the sections (11), which are detached from the sample (10) are caused to float on a liquid surface and are thereafter transferred onto a solid carrier element (20). For at least for some of the sections (11) detached from the sample (10) a position and an orientation on the solid carrier element (20) are determined by monitoring the placement of these sections (11) onto the solid carrier element (20) using a monitoring system (400) comprising a camera (410), obtaining monitoring data. A method (2000) for the three-dimensional reconstruction of a microscopic sample (10), a microtome system and a computer program are also part of the present invention.

Abstract

The present invention concerns a method and a system for imaging at least a part of a specimen by means of two microscopy imaging methods, where a surface (11) of the specimen (10) is imaged by means of a first microscopy imaging method, where a replica (25) of the surface (11) to be imaged by means of the first microscopy imaging method is produced and this replica (25) is simultaneously imaged by means of a second microscopy imaging method, where the images produced by means of the first and the second microscopy imaging methods are overlaid at the correct scale.

Abstract

A method including one or more steps of a procedure for preparing a microscopic sample for a high-pressure freezing process is provided, wherein the sample is provided using an arrangement comprising a middle plate of a high-pressure freezing cartridge and an incubation chamber, wherein the middle plate is attached to a lower surface of a bottom of the incubation chamber by an adhesive and detachable from the incubation chamber by effecting a relative movement between the middle plate and the incubation chamber, wherein the sample is provided on an enclosing element which is fitted into an opening of the middle plate, and wherein, when the sample is to be subjected to the high-pressure freezing process, the relative movement between the middle plate and the incubation chamber is effected, thereby detaching the middle plate with the sample from the incubation chamber. Means to implement the method are also part of the present disclosure.

Abstract

A computerized model compatibility regulation method for imaging applications first performs a target domain B application by computing means using at least one image X and target domain B image analytics to generate a target domain B application output for X. The method then applies a reference domain A application by computing means to generate reference domain A application output for X. The method further performs a compatibility assessment to generate at least one compatibility result for X. In addition, the method checks the compatibility result for X and if the check output is incompatible, the method performs online correction to generate a corrected application output for X.

Abstract

A system for identifying presence of a conjugate in an image comprising one or more processors and one or more storage devices is provided. The system is configured to acquire spectral data associated with a sample to be imaged. Dispersion coefficients are optimized for acquired spectral data. A corrective phase function is calculated using the optimized dispersion coefficients. A negative corrective phase function is applied to a signal to provide a resulting image. It is determined if the resulting image has degraded signal strength or an enhanced signal strength relative to an original image. A reference arm shift is calculated if it is determined that the resulting image has enhanced signal strength. A position of a reference arm of the system is adjusted based on the calculated reference arm shift to move a conjugate image out of view.

Abstract

A system for optimizing optics is provided. The system is configured to calibrate a position of a reference arm of an interferometric imaging system such that an image of a sample is visible when the sample is positioned at a working distance of an objective lens to provide an initial calibrated position. An image is obtained using the initial calibrated position. Image quality of the obtained image is assessed to determine if the obtained image is a valid image. A path length of the reference arm is adjusted if it is determined that the obtained image is not a valid image. A difference between the calibrated position of the reference arm and the adjusted position of the reference arm is calculated. System elements are adjusted based on the calculated difference such that the sample is visible when the sample is positioned at the working distance at the adjusted position.

Abstract

Systems for determining an apex of curvature In an image, obtained from, a sample are provided. The systems include -an imaging system configured to obtain a plurality of scans of a sample using a radial pattern; and a processor associated with the imaging system. The processor is configured to segment and curve fit each of the plurality of scans to a surface of the sample; determine an apex.for each curve associated with each of the plurality of scans; determine a true apex, among all determined apexes using a derivative of least value; calculate an XY offset based on the determined true apex; map the true apex to an origin where X and Y are equal to zero; and adjust the coordinates associated with remaining apexes not determined to be the true apex based on the calculated offset.

Abstract

A method is proposed for generating a series of sections of a microscopic sample (10), in which the sections are detached from the sample (10) by means of a blade (21), collected, and placed onto a transfer device. A sequence in which the sections are detached from the sample is detected; detachment of the sections from the sample (10) and/or collection of the sections and/or placement of the sections onto the transfer device is monitored by means of one or several observation cameras, accompanied by the acquisition of moving-image data; the sections are tracked in the moving-image data; and positions of the sections on the transfer device are correlated, on the basis of the tracking in the moving-image data, with the sequence in which they were detached from the sample. Also subjects of the present invention are a method for three-dimensional reconstruction of a microscope sample (10), in which sections of the sample (10) are microscopically investigated accompanied by acquisition of section image data and in which the section image data are assembled into a volume image; and a corresponding microtome system (100) having a microtome (110) and a data acquisition and evaluation unit (120), and having one or several observation cameras (30).
