Abstract

The present invention is in the general area of post-harvest fruit disease detection, more pertinently the present invention is directed towards methods for colorimetric detection of pathogenic fungi in their latent phase in harvested fruit, using a paper/plate -based reverse transcription loop- mediated isothermal amplification (RT-LAMP). The present invention provides a paper-based device for performing reverse transcription loop-mediated isothermal amplification (RT-LAMP) with real-time simultaneous detection of C. gloeosporioides latent infections in tomatoes. RT- LAMP -based PAD platform comprises a paper substrate on which the RNA amplification reaction occurs. The assay provided by the present invention is highly specific for the latent stage of C. gloeosporioides with an analytical sensitivity of 0.5 pg of total extracted RNA, with results obtained within 40 min and hence can be efficiently employed for identifying C. gloeosporioides in resource-limited setting.

Abstract

A method and system for hyperspectral imaging uses a flock of at least two sensors which provide images of an overlapped region-of-interest in at least two different spectral bands. The latter may include a visual band and one or more infrared bands. Using a variety of calibration, registration, super-resolution, and data fusion algorithms, a signal processor corrects and analyzes the images to form a hyperspectral image cube with high spatial resolution. The signal processor typically is in communication with or integrated with a tracking station, and it may be spaceborne, airborne, shipborne, or gound-based. An exemplary neural network is provided for implementation of the method for hyperspectral imaging.

Abstract

A method and system for early detection of Bovine Mastitis (BM) uses enhanced chemiluminescence (CL) to determine Haptoglobin (Hp) levels in highly diluted milk samples. The samples are placed in the wells of one or more bio-functionalized Hemoglobin (Hb) modified CL assay plates. Hp-Hb binding occurs in those samples containing Hp. After a first pre-determined time duration, the wells are treated with a CL solution containing luminol and peroxide, and a colloidal suspension of crosslinked nanoparticles. After a second pre-determined time duration, the CL intensity approaches a steady state value. A processor analyzes CL images provided by a camera in order to measure CL intensity and to determine an estimate of Hp level in each well, based on a pre-determined regression curve. The processor then forms a BM clinical diagnosis by combining the estimated HP levels from wells with different milk sample dilutions.

Abstract

A label-free optical biosensing system and method provide high sensitivity and specificity for in situ detection and activity estimation of serotypes, such as Botulinum Neurotoxins (BoNT). Pre-fabricated thin-film support structures are treated with a competitive immunoassay coupled to a biochemical cascade reaction, which provides optical signal amplification. When the thin-film support structures receive a target analyte and are exposed to polychromatic light, reaction products cause a change in average refractive index which appears in reflectivity spectra measured by an optical interferometer. Optical signal amplification enables a linear response for serotype concentrations of only a few picograms per millilitre, as well as a level-of-detection threshold of 5.0 picograms per millilitre or less. The specificity and selectivity of the method have been verified in studies using various combinations of different serotypes as a target analyte. Similarly, the serotype activity is estimated by an adjunct sensing platform.

Abstract

The present application discloses a method for the early detection of bacterial diseases of potato tubers. More particularly, the present application discloses a method for identifying bacteria which cause blackleg and tuber soft rot diseases in potato tubers based on the identification of unique patterns of volatile compounds. The method disclosed herein can serve for the differentiation of non-inoculated samples from inoculated samples, distinguishing and identifying inoculation by different bacterial genera (Pectobacterium and Dickeya spp), different bacterial species and different bacterial sub-species. The detection relies on specific combinations of volatile compounds associated with each of the above-mentioned groups, which can serve as an applicable tool to detect tuber pathologies in storage rooms and shipments.
