Abstract

An apparatus for generating ions from sample material deposited on a sample carrier includes a chamber that keeps the sample material in a conditioned environment, an ablation device that locally ablates the deposited sample material and converts it to the gas phase, an energy input device that exposes the ablated sample material to an input energy burst, and an extraction device that extracts ions from the ablated sample material exposed to the input energy burst and transfers them to an ion processing apparatus, the ablation energy burst and/or input energy burst comprising a beam of coherent electromagnetic waves and the ablation device and/or the energy input device comprising a beam distributor that provides a plurality of optically parallel beam paths which each have a frequency multiplier unit and are combined again, wherein all frequency multiplier units emit a uniform output frequency. Abstract

The invention relates to a method for separating ions and a trapped ion mobility separator. The trapped ion mobility separator comprises an ion guide extending in a closed loop which contains a gas substantially at rest through which ions pass. An axial force acting on the ions is provided, being caused by an electric field barrier providing an electric field gradient that moves around the ion guide, and having an effect on the movement of the ions that is dependent on ion mobility. At least one operating parameter of the trapped ion mobility separator, which has an impact on the mobility separation, is varied as a function of time, so that the ions are pushed controlled along the moving electric field barrier towards a high potential end of the electric field barrier, wherein the ions are ejected laterally from the ion guide before slipping over said electric field barrier.

Abstract

The invention proposes preparing biological samples for spectrometry which contain cell structures and/or whole cells of human or animal origin (e.g. thin human and animal tissue sections) or prokaryotes (e.g. microorganisms), and which require constant relative humidity, in a temperature-controlled gas volume whose humidity is determined by a saturated substance solution, for example a suitable salt solution. The invention exploits a physico-chemical phenomenon called “deliquescence”, which manifests itself by keeping the relative humidity above the saturated substance solution constant with a high degree of precision when a specified temperature is maintained. Pure succinic acid exhibits deliquescence at approx. 99% relative humidity, for example. Since an enormous variety of deliquescent salts and other suitable substances are available, it is possible to find the suitable substance for almost any desired relative humidity, with adjustment of the temperature, where necessary.

Abstract

The invention relates to the preparation of biological cells for the mass spectrometric analysis of cellular properties such as taxonomic classification, antibiotic resistances, response to drugs or other active substances, and others. The cells can be prokaryotic or eukaryotic microorganisms which have particularly been cultivated directly on a mass spectrometric sample support, or eukaryotic cells from tissues or cell cultures. The invention proposes that the cells are not disrupted by adding matrix solution for a subsequent ionization by matrix-assisted laser desorption (MALDI), but that they are disrupted in a separate treatment step using acids and/or solvents on the sample support itself. Surprisingly, the cell proteins released then adhere to the sample support so that they can be carefully washed with buffer solution to remove salts and other soluble impurities which can stem from earlier treatment steps, for example from nutrient solution.

An ion mobility separator includes an ion path with a central axis along which ions travel, the ion path containing a gas. Abstract

The invention proposes preparing biological samples for spectrometry which contain cell structures and/or whole cells of human or animal origin (e.g. thin human and animal tissue sections) or prokaryotes (e.g. microorganisms), and which require constant relative humidity, in a temperature-controlled gas volume whose humidity is determined by a saturated substance solution, for example a suitable salt solution. The invention exploits a physico-chemical phenomenon called “deliquescence”, which manifests itself by keeping the relative humidity above the saturated substance solution constant with a high degree of precision when a specified temperature is maintained. Pure succinic acid exhibits deliquescence at approx. 99% relative humidity, for example. Since an enormous variety of deliquescent salts and other suitable substances are available, it is possible to find the suitable substance for almost any desired relative humidity, with adjustment of the temperature, where necessary.

The invention relates to the preparation of samples for mass spectrometric analyses with matrix-assisted ionization (e.g. MALDI), particularly using the matrix material 2,5-dihydroxyacetophenone (DHAP), and comprises sample preparation in microvessels with the addition of beads or other solid bodies. In this way, particularly thorough mixing can be achieved by shaking (“vortexing”) the microvessels, on the one hand, while crystallization of the matrix material can be initiated in the liquid, on the other hand. It is sufficient to have around 5 to 15 glass beads, 0.5 millimeters in diameter, which scrape against each other and against the vessel wall, and thus cause a DHAP solution to start becoming turbid after one to two minutes by forming tiny crystal nuclei. This allows simultaneous, fully automated preparation of, in particular, a large number of samples, including pipetting onto sample support plates by means of a pipetting and shaking robot.

Abstract

The invention relates to imaging mass spectrometry on thin sample sections, in particular using MALDI, where a high lateral image resolution means that a plethora of mass spectra has to be acquired and the image acquisition runs over many hours. The quality of the mass spectra deteriorates considerably over time in such cases. The invention is based on the finding that the decrease in spectral quality of continuous measurement series over many hours is only partially caused by a decrease in detector gain, and that another significant cause is a decrease in the number of usable ions per ion generating pulse, which is attributable to several phenomena that are difficult to regulate. The invention now proposes to instead regulate only the detector gain, and such that not only the decrease in the detector gain is compensated, but also the decrease in the number of usable ions per ion generating pulse.

Abstract

The invention relates to the operation of an energy-focusing and solid-angle-focusing reflector for time-of-flight mass spectrometers with pulsed ion acceleration into a flight tube, e.g. from an ion source with ionization by matrix-assisted laser desorption (MALDI). The objective of the invention is to generate high mass resolution in wide mass ranges up to high masses above eight kilodaltons by varying at least one operating voltage on one of the diaphragms of the reflector which can be varied according to a suitable time function during the spectrum acquisition. It may also be advantageous to adapt the operation of the accelerating voltages in the starting region of the ions accordingly. These measures make it possible to achieve a mass resolution much higher than R=100,000 in a wide mass range extending up to and above eight kilodaltons.
