Abstract

Ultrasound transducer element arrays using acoustic ensonification drive patterns via a patient interface for sonosensitizer activation in sonodynamic therapy. Incoherent acoustic field generation varying phase, frequencies, and/or amplitude via controlled delivery of low intensity planar acoustic waves. Method includes generating a first and a second signal to generate respective acoustic ensonification drive patterns with phase, frequency, and amplitude and generating at least one relative phase, frequency or amplitude difference to generate a third incoherent acoustic ensonification pattern to activate a sonosensitizer. Calibration and complementary therapy procedures to improve cell susceptibility of cells to sonodynamic therapy are disclosed.

Abstract

Ultrasound transducer arrays are provided to for example, initiate and enhance therapeutic treatments with a normalized, randomized, and/or incoherent acoustic pressure field. Optimizing pressure uniformity (peak and average pressures) throughout the incoherent pressure field, and maximizing volume of that incoherent field may be achieved through controller filtering steps to assign unique element waveform phases for each element in the array. Ultrasound may be used alone, to activate a drug, pro drug, sonosensitizer, and/or microbubble additives, and can be combined with other energy (e.g., radiation, magnetism), for treatments including cancer, neurological disease, mood condition, sleep apnea, inflammation, and/or orthopedic diseases, and opening the blood brain barrier to improve access to the drugs and additives. Ultrasound transducer systems may used with a cooling system, an alignment device a monitoring system an authorization system (e.g., identification bar code, key) and/or a treatment planning system.

Abstract

Methods of treating tumors by administering compounds to a patient are provided. Compounds such as drugs, may be administered to the patient orally, by injection, intravenously, or topically, which then accumulate preferentially as compounds such as protoporphyrin IX (PpIX) in tumor cells. After such accumulation, compounds such as PpIX are then activated in various aspects to treat tumors cells, thereby treating cancer. Cancers such as glioblastoma may be treated.

Abstract

Disclosed are methods of obtaining zero vergence ultrasound waves for providing sonodynamic therapy with ultrasound waves. The method includes coupling a sonodynamic therapy device with an array of flat piezoelectric transducers to a skin surface. A controller is configured to generate an electrical drive signal at a frequency, modulate the drive signal, and drive the transducer with the modulated drive signal at the frequency to produce a zero vergence ultrasound wave to produce an average acoustic intensity sufficient to activate a sonosensitizer in a treatment region without damaging healthy cells in the treatment region.

Abstract

A catheter is disclosed that may be used in a minimally invasive internal treatment (e.g., sonodynamic therapy). The catheter can include a housing, a portion of which may be positioned in contact with internal tissue of a patient during a minimally invasive sonodynamic or photo-sonodynamic therapy procedure. The catheter may include multiple electrically independent ultrasound transducers. The ultrasound transducers can be configured to emit ultrasound energy into the internal tissue of the patient. The ultrasound energy that is emitted from the catheter may reach a target tissue depth at a relatively low temporal average intensity (e.g., less than 50 W/cm2). Such ultrasound energy may activate the sensitizer.

Abstract

An intracranial treatment apparatus comprises an outer shaft having a proximal end and a distal end for positioning within the tissue region of the brain. The outer shaft defines a lumen extending between the proximal end and the distal end of the outer shaft and having at least one aperture adjacent the distal end of the outer shaft. An inner light-delivery element having a distal end and a proximal end is adapted to be operatively connected to the light source. The light-delivery element is configured to be received within the lumen and extend from the proximal end of the shaft to adjacent the distal end of the shaft. The light-delivery element is adapted to deliver light from the light source through the at least one aperture of the outer shaft to the tissue region of the brain in proximity to the distal end of the outer shaft.
