Abstract
Lung infections remain a substantial concern, even in wealthy countries. For instance, the emergence of a new betacoronavirus SARS-CoV-2 has led to a major health-related crisis associated with a significant mortality in intensive care units, due to the pulmonary complications of COVID-19. The inventors aim at developing a new therapeutic approach based on the local delivery of apolipoprotein A1 nanoparticles into the lung by aerosolization. These particles may display antiviral properties that could be improved by loading them with small interference RNAs specifically targeting SARS-CoV2 genome and antiviral drugs (such as the hydrophobic chloroquine). Their preliminary results have shown in preclinical mouse models that aerosolization allowed apoA1 nanoparticle uptake and persistence (up to 72h) in pulmonary cells. In addition to the potential intrinsic antiviral activity of apoA1, these particle display important anti-inflammatory effects that may limit the excessive immuno-inflammatory host response that is deleterious for the lung. Thus, the present invention relates to aerosolization of HDL for the treatment of lung infections.

Abstract
The present invention relates to the field of vaccination against flaviviruses, and in particular against Zika virus. The invention further relates to methods for producing or selecting antibodies directed against said flaviviruses. The invention further relates to isolated polypeptides, recombinant cells, and attenuated viruses, suitable for eliciting an immune response. The inventors have shown that a 20-mer peptide GSQHSGMTVNDIGYETDENR representing the glycan-loop (GL) of the Zika virus (ZIKV) strain MR766 is antigenic in a peptide-based ELISA format. To the knowledge of the inventors, this is the first report demonstrating the antigenicity of a fragment of the Envelope protein of a flavivirus, within Domain I (E-DI), which explicitly includes the glycan-loop itself.

Abstract
Flaviviruses are largely pathogenic to humans and other mammals and mostly are transmitted by mosquitoes. To combat flavivirus infections, it is urgent to propose safe and effective anti-viral compounds not only to impair the virus spread but also to mitigate the associated morbidities. Despite the development of antiviral agents from synthetic sources, a novel antiviral approach (economical, simple and environmentally friendly) is to use natural sources such as nutraceutical as preventive treatments against viral infections. The inventors demonstrated that flavonoid isoquercitrin exerts antiviral activity against African historical and Asian epidemic strains of ZIKV in human hepatoma, epithelial and neuroblastoma cell lines. They showed that isoquercitrin acts on ZIKV entry by preventing the internalization of virus particles into the host cell. Next the inventors demonstrated that Q3G exert antiviral activity against Yellow Fever Virus (YFV). These results suggest that Q3G represents a potential prophylactic agent targeting the entry of two medically relevant flavivirus and could thus be used to prevent flavivirus infection. Accordingly, the the present invention relates to use of quercetin-3 -O-glucoside for the treatment of flavivirus infections.

The present invention relates to vaccine compositions comprising an attenuated mutant Zika virus. The inventors have introduced mutations at very specific positions that abrogate the N-glycosylation site on the E protein of the epidemic strain which will prevent the generation of auto-antibodies responsible for Guillain-Barre syndrome. The inventors have also produced additional mutations of the virus that result to a dramatic reduction of the cytopathic effects without affecting the capacity to produce high titers of virus. In particular, the present invention relates to an attenuated mutant Zika virus comprising a protein E of the epidemic strain wherein at least one amino acid residue at position 152, 156 or 158 is mutated.

Abstract
The present invention relates to a genomic sequences encoding for an attenuated mutant Zika virus. The inventors have introduced some specific substitutions at very specific positions in the epidemic genomic sequence for restoring some fixation sites for miR-4279 that were originally present in the endemic genomic sequence. Moreover the inventors have additionally introduced mutation leading to the abrogation of the N-glycosylation site on the E protein which will prevent the generation of auto-antibodies responsible for Guillain-Barre syndrome. The inventors have produced additional mutations of the virus that result to a dramatic reduction of the cytopathic effects without affecting the capacity to produce high titers of virus. In particular the present invention relates to a genomic sequence characterized by the sequence represented by SEQ ID NO:1 wherein at least one site of fixation for miR-4279 is restored.

Abstract
The present invention is related to a therapeutic mucosal or intradermal or intraepithelial vaccine composition for use in a method for treating an infection by a HIV virus in a human in need thereof, comprising: - an antigen made of inactivated autologous HIV virus; and - a tolerogenic adjuvant which, when combined to said antigen in said therapeutic vaccine composition, induces and, preferably, maintains immunotolerance to said HIV virus in said human, thereby treating said infection in said human. More precisely, the therapeutic vaccine compositions of the present invention produce a virus-specific non-cytotoxic MHC-lb/E-restricted suppressive CD8+ T cell- induced immunotolerance ("Ts" immunotolerance for "T suppressive" immunotolerance) upon mucosal or intradermal or intraepithelial administration to humans. "Ts" immunotolerance is associated with the suppression of viral replication. The present invention also relates to a method for treating an infection by a HIV virus in a human in need thereof, comprising at least the step of mucosally or intradermally or intraepithelial administering a therapeutic vaccine composition as defined above to said human.
