Abstract

The present invention discloses a novel class of covalent small molecules comprising 2-amino benzophenone derivatives for targeted ferroptosis induction in cancer cells. The present invention also includes strategies for target identification, hit-to-lead optimization, and combination therapies to enhance therapeutic efficacy. A synthesis and characterization of a library of unique organic structural scaffolds encompassing natural/unnatural amino acids, aromatic and heterocyclic linkers coupled with electrophilic units possessing diverse nucleophile reactivity profiles is provided. The molecules exhibit sub-micromolar to nanomolar inhibitory potency (IC50) when screened against a panel of cancer cell lines including MCF7 (breast cancer), MDA-MB-231 (triple negative breast cancer), 22Rv1 (Prostate), PC3, (prostate cancer), Jurkat J6 (acute T-cell Leukaemia), RPMI 8226 (B lymphocytes), U87MG (glioblastoma), HCT-116 (colon cancer) and A375 (melanoma)) and kidney cells (HEK293). The molecules exhibit potent antiproliferative effects across various cancer cell lines, offering a promising alternative to conventional chemotherapies with reduced side effects.

Abstract

The present invention provides modified adeno-associated virus (AAV) vectors designed for gene therapy for treating hemophilia B The modified AAV vectors are obtained with enhanced transduction efficiency and immune evasion capabilities achieved through precise modifications at post-translational modification (PTM) sites on AAV8 and AAVrh10 capsids. The modified AAV vectors offer advantages including improved therapeutic efficacy at lower vector doses, similar immunogenicity profile as wild type AAV vectors, and versatility for treating hemophilia B and similar liver disorders (e.g., Gaucher's disease). The present invention further encompasses a method for manufacturing the modified AAV vectors, ensuring their precise and reliable production. The modified AAV vectors has potential for long-term gene expression and adaptability for other genetic disorders which makes the present invention a significant advancement in the field of gene therapy.

A system and a method for terminating ransomware based on detection of anomalous data is disclosed. The system and the method comprise a registry activity monitoring subsystem, a file trap monitoring subsystem, a decision generating subsystem, and a termination subsystem. The registry activity monitoring subsystem is configured to generate first data and the file trap monitoring subsystem is configured to generate second data. The first data and the second data are transferred to the decision generating subsystem for retrieving Process IDs (PIDs) to initiate a ransomware termination process. Upon confirmation from the decision generating subsystem, the termination subsystem is configured to terminate the retrieved Process IDs (PIDs) to terminate the ransomware based on the detection of the anomalous data from one or more computing devices.

Abstract

A system and method for kernel-level active darknet monitoring in a communication network is disclosed. The system receives TCP packets from initiator devices and determines dark internet protocol (IP) addresses in the packets by comparing destination IP address with a plurality of IP addresses stored in a dark IP pool. If the packets are determined to be destined for dark IP addresses, the system modifies flag and type in socket buffer and routing entry associated with packets. The system retrieves outbound interface IP addresses and creates a socket corresponding to the retrieved outbound interface IP addresses, for transmitting a SYN-ACK packet corresponding to the TCP packets. The system is configured to monitor dark IP addresses received through network traffic. Finally, the system outputs the SYN-ACK packet to initiator devices to establish a three-way handshake. The system receives an ACK from initiator devices (attackers) to establish a 3-way handshake.

A haptic tactile smart watch for the visually impaired is provided. The tactile smart watch comprises a watch case having a tactile digital interface. One or more tactile push buttons are configured on an outer circumference of the watch case. The one or more tactile push buttons activate one or more functions associated with the tactile smart watch. One or more touch sensitive indicators receive one or more touch inputs from a user input. An optimized AAV vector for gene therapy of muscular dystrophy is provided. The optimized AAV vector includes a plurality of mutant AAV9 vectors and a microdystrophin transgene (p.AAV-CBA-kozak-μDys). The optimized AAV vector demonstrate increased transduction efficiency, gene expression levels, and can potentially achieve optimal therapeutic efficacy in humans at lower vector doses. The optimized AAV vector also demonstrate improved transduction and dystrophin gene expression in a mice model of Duchenne muscular dystrophy.

A formulation and method for treating prostate cancer is provided. Present invention relates to a rapid soil testing device designed for measuring the soil physical and chemical properties. Techniques for determining presence of cancerous cells in breasts of a subject are described. In an example, transient temperature response data for the breasts is received. The transient temperature response data is then converted to steady state temperature response data. A set of features for the breasts are then extracted based on the steady state temperature response data. An environmentally benign method for producing binary microfabrication masks is disclosed. An optical target may be provided that includes a water-soluble polymer material in contact with an ultraviolet radiation transmittable substrate. A laser may be focused on a primary mask to produce a mask image, the mask image thereafter being reduced by demagnification optics to provide a reduced image. A thin film transistor (TFT) includes a hole transport layer having a first side and a second side and an electron transport layer having a first side and a second side. The first side of the electron transport layer is directly interfaced to the second side of the hole transport layer. The electron transport layer includes a material having greater ionization potential and greater electron affinity than the hole transport layer, thereby forming a hole barrier and an electron barrier at the junction between the electron transport layer and the hole transport layer. A channel in the TFT is created by current injected into the electron transport layer from a gate electrode rather than by an electrostatic field generated by voltage applied to the gate electrode. Abstract

A dual reporter plasmid construct for measuring Bone Morphogenetic Proteins (BMP) Signalling is provided. The dual reporter construct includes a BMP Responsive element of Sequence ID No 1 a minimal promoter gene cloned downstream of the BMP responsive element, a firefly luciferase reporter gene of Sequence ID No 3 cloned downstream of the BMP responsive element and the first minimal promoter gene, a constitutive promoter cloned downstream of the firefly luciferase reporter gene, a renilla luciferase reporter gene of Sequence ID No 7 cloned downstream of the constitutive promoter. The renilla luciferase reporter gene acts as an internal control that normalises any altered activity of the firefly luciferase gene in presence or absence of BMP or BMP agonist. The firefly luciferase reporter gene activity provides a quantitative measure of activity of the BMP responsive element.

Abstract

Methods and devices for preparing microscale polymer relief structures from a thin polymer layer on an absorbing substrate are described. The described methods are ultrafast (about 8 nanoseconds) and allow formation of patterned microstructures having complex morphologies and narrow line widths that are an order of magnitude smaller than the masks used in the methods.

Abstract

Disclosed herein are biodegradable hydrogel scaffolds for use in tissue engineering. The hydrogel scaffolds are composed of synthetic terpolymers complexed with polyvinyl alcohol (PVA), which facilitate cell-sheet and tissue growth. In the presence of a monosaccharide, the PVA-hydrogel is dissolved and cell-sheets are released for harvesting. Further disclosed herein are methods for producing PVA hydrogels which support tissue growth. Tissue engineering applications and methods are also disclosed.

Abstract

Drive signals for a display device may be generated using Separable Nonnegative Matrix Series Representation (SNMSR) of source image data and applying a non- negative matrix factorization (NNMF) process to source image data to generate approximation image data (Ii), partial sum image data (Pi) and residue image data (Ji). Iteratively, NNMF may be applied to Ji such that subsequent Ii and Ji may be generated, where each Ii can be associated with a corresponding sub-frame image. At each iteration, the Ii may be sent to the display buffer for selective activation of multiple row and column drivers during a single sub-frame interval. At each iteration, a determination may be made if a predetermined criterion is satisfied. The iterations may be terminated and the series truncated when the predetermined criterion is satisfied. Integration of the sub-frame images displayed over a complete frame interval by human eye effectively corresponds to the source image.

Systems and methods for imaging characteristics of a sample and for identifying regions of damage in the sample are generally described. Some example systems and methods for non-destructive evaluation of regions of material may operate in a direct current (DC) mode in which the system directly images regions of material where weak structural damage has occurred by imaging a self magnetic field generated by a DC electric current coupled through the material. Abstract

A method, system and device are described for generating high-resolution flow-field data from sparse measurement data by interpolating from a reference database of pre-computed, highly-resolved fluid dynamic simulation results, to generate a highly-resolved set of flow field data which correspond to the sparse measurements. In one embodiment, the method, system and device use Principal Component Analysis techniques to interpolate the multidimensional fluid flow data. Applications of the invention are, amongst others, in the field of biomedical diagnosis involving fluid dynamic modeling of biological fluid systems such as blood flow in an artery.

The formation of self-assembled patterns and their modulation in a visco-elastic surface layer via substantially elastic deformations induced by displacement of a stamp in contact with the surface layer are disclosed. Methods and apparatus of the present invention may be employed to form arrays of micrometer and sub-micrometer channels, wells and pillars and other complex structures by using displacements of the single stamp. The elastic structures thus formed can be altered in-situ and also preserved, if desired.
