Abstract

Method and systems are provided for defining test sample read count limits for a range of microbial sequence reads. A computing device may generate a stabilization framework usable to determine a viability of a stabilization solution to stabilize a particular microbial species. The stabilization framework may include a first read count limit defined based on an enumerated abundance of the microbial species in a control sample multiplied by a first sequencing range value. The computing device may receive a read count value corresponding to an enumerated abundance of the microbial species in a mixture comprising a biological sample and the stabilization solution. The stabilization framework maybe applied to the read count value to generate an output based on whether the read count value exceeds the first read count limit. The computing device may then select, the stabilizing solution for use with subsequent biological samples.

The present invention generally relates to a sample receiving device for releasably storing a substance. Abstract

The disclosure relates to devices, solutions and methods for collecting and processing samples of bodily fluids containing cells (as well as embodiments for the collection, and processing and/or analysis of other fluids including toxic and/or hazardous substances/fluids). In addition, the disclosure relates generally to function genomic studies and to the isolation and preservation of cells from saliva and other bodily fluids (e.g., urine), for cellular analysis. With respect to devices for collection of bodily fluids, some embodiments include two mating bodies, a cap and a tube (for example), where, in some embodiments, the cap includes a closed interior space for holding a sample preservative solution and mates with the tube to constitute the (closed) sample collection device. Upon mating, the preservation solution flows into the closed interior space to preserve cells in the bodily fluid. The tube is configured to receive a donor sample of bodily fluid (e.g., saliva, urine), which can then be subjected to processing to extract a plurality of cells. The plurality of cells can be further processed to isolate one and/or another cell type therefrom. The plurality of cells, as well as the isolated cell type(s), can be analyzed for functional genomic and epigenetic studies, as well as biomarker discovery.

Abstract

Methods, compositions, and kits for stabilizing both human and microbial deoxyribonucleic acid (DNA) present in complex biological samples, such as feces, are disclosed. In particular, aqueous compositions for stabilizing DNA contained in biological samples at ambient temperature are disclosed, together with associated methods and kits using same. In one aspect, the compositions comprise a chelating agent present at a concentration of at least about 150 m M, and the composition has a pH of at least about 9.5.

Abstract

The present application provides a method and aqueous composition for stabilizing and isolating nucleic acid from whole blood or blood fraction samples. The method and aqueous composition can be used to stabilize and isolate nucleic acid from whole blood, plasma, buffy coat or total white blood cell isolates. The aqueous composition can comprise a denaturing agent, a chelating agent and a buffering agent, preferably in the absence of a chaotrope, and at a pH greater than 5.0. The samples can be stored at varying temperatures, preferably at room temperature, and for prolonged lengths of time while maintaining integrity of the nucleic acid.

Abstract

The disclosure relates to devices, solutions and methods for collecting and processing samples of bodily fluids containing cells (as well as embodiments for the collection, and processing and/or analysis of other fluids including toxic and/or hazardous substances/fluids). In addition, the disclosure relates generally to function genomic studies and to the isolation and preservation of cells from saliva and other bodily fluids (e.g., urine), for cellular analysis. With respect to devices for collection of bodily fluids, some embodiments include two mating bodies, a cap and a tube (for example), where, in some embodiments, the cap includes a closed interior space for holding a sample preservative solution and mates with the tube to constitute the (closed) sample collection device. Upon mating, the preservation solution flows into the closed interior space to preserve cells in the bodily fluid. The tube is configured to receive a donor sample of bodily fluid (e.g., saliva, urine), which can then be subjected to processing to extract a plurality of cells. The plurality of cells can be further processed to isolate one and/or another cell type therefrom. The plurality of cells, as well as the isolated cell type(s), can be analyzed for functional genomic and epigenetic studies, as well as biomarker discovery.

Abstract

A closure for a container has a closure body having a first cylindrical portion and a second cylindrical portion opposite the first portion. The closure also has a connector disposed in at least one of the first and second portions. The connector is adapted to connect one of a sample collection device and an applicator to the closure. A containing apparatus including the closure, a container, and optionally one of a sample collection device and an applicator is also disclosed. A method of using the containing apparatus is also disclosed.

Abstract

The present invention generally relates to a sample receiving device for releasably storing a substance. The samplereceiving device includes a lid having a reservoir for retaining the substance, and a pierceable barrier sealing the substance withinthe reservoir; and b) a funnel for receiving a sample and configured for closure by the lid. The funnel is configured for releasableattachment to a sample receptacle such that a sample can be provided to the funnel and travel through the channel in the funnelinto the sample receptacle. Further, the funnel includes one or more cutting ribs for cutting the pierceable barrier such that uponcutting of the pierceable barrier the substance is released from the reservoir, flows through the channel in the funnel and into thesample receptacle to be mixed with the sample. Abstract
