Abstract

Transition metal complexes as catalysts and, occasionally, basic pH 9-11 as a reaction condition were employed to imitate the catecholase activity. Using base as a catalyst, under photo- mediated condition the catalytic aerobic oxidation of the proxy substrate 3,5-di-tert- butylcatechol (DTBC) in polar protic solvent very fast was shown in this invention to be a functional mimic of catecholase activity. The Michaelis-Menten kinetic model's enzymatic substrate inhibition pattern was used to describe the kinetic manifestation of 3,5-di-tert- butylcatechol (DTBC) oxidation. Under photo-mediated condition the oxidation of 3,5-di-tert- butylcatechol (DTBC) was observed very fast which may be due to the higher energy increases the activation energy and under basic pH condition catechol oxidizes to 3,5-di-tert- butylcyclohexa-3,5-diene-1,2-dione (DTBQ).

Abstract

The invention relates to chiral amino acids as well as symmetrical diamino triazine substituted phthalazinones as Poly(ADP-ribose)polymerase (PARP) Inhibitors, process for the preparation and uses thereof as potential 5 anticancer agents. The invention particularly relates to a process for the preparation of symmetrical for new. The compounds inhibited the growth and development of BRCA2 mutant cells with an IC50 value of the most active compounds in the range of 2-57 nM. The active compounds selectively induce cytotoxicity in BRCA2 mutant cancer cells compared to wild type cells with a mutant fold selectivity in range of 112- 2500-fold. The active compounds are capable of killing the BRCA mutated cancerous cells and show significant PARP 10 activity as well as show low cytotoxicity to wild type cells. Most importantly, our hit molecules demonstrated approximately 50% or greater cell viability of mice HSPCs. This suggests an improved hematological safety profile of our hits, distinguishing them from FDA-approved PARP inhibitor Olaparib.

Abstract

The present invention relates to a mutant LC3B allosteric switch, wherein three specific residues have been mutated to either reduce or enhance receptor binding, compared to the wild- type protein. In particular, the present invention provides a method for generating conformational ensemble of lipidated protein on the membrane and introducing specific mutations within the protein activation switch that is indirectly altering protein binding pocket without disrupting its functional domains. The identification of an activation site opens avenues for the development of small molecules or peptides to enhance or regulate protein activity, providing a new dimension for therapeutic interventions. Therefore, by linking the structural insights of lipidated forms to functional outcomes, the invention advances knowledge on LC3 activity regulation, which could further be utilized to modulate the autophagy pathways.

Abstract

This invention provides a novel expedient one step synthetic procedure for the preparation of derivatives of dehydrocostus lactone and costunolide using diverse piperazine under very mild reaction condition and short period of time. The synthesized derivatives have improved safety profile along with anti-leishmanial killing potential. The potent leads displayed significant leishmanicidal potency and satisfactory selectivity against major macrophage cell lines (J774.A1 and RAW 264.7). Further, ex vivo analysis validated their potential for the clearance of intracellular amastigote burden with a better anti-amastigote selectivity index. The potent hits 1po and 2pp caused alteration in structural morphology, intracellular ROS generations, depletion in ATP pool indicating mitochondrial disfunction, accumulation of lipid inclusions, and disruption of sterol membrane. This study confirmed efficacy of synthesised derivatives against L. donovani supported by exploration of mechanistic insight. Further exploration of potent derivatives may lead to development of novel drug candidates to fight against parasitic infections.

Abstract

A filtration unit assembly (100) to separate solid and liquid mixtures is disclosed. A mixture of a solid and a liquid enters the top dish (110) through one or more inlets provided on the top dish. The received mixture for filtration then passes through a rotating drum (140) that is housed within a shell (150). The mixture of the solid and liquid enters the rotating drum and is exposed to a screw action by the rotary motion of the rotating drum (140). The resulting screw action separates the solid and the liquid as the liquid gets sucked towards the axis while the solids are pushed away from the axis of rotation of the rotating drum. The rotating drum (140) may be provided with perforations to aid in separation. A system for filtration that includes a filtration unit assembly (100) is also disclosed.

Abstract

The present invention relates to a recombinant vector for genome editing in rice. The vector comprises of GluB1 promoter of rice along with Cas 12 a gene and a guide RNA. Editing the targeted gene within localized tissue effectively mitigates the pleiotropic effect observed in non- targeted tissues, thus reducing the potential for unintended consequences. Tissue-specific genome editing (TSGE) plays a pivotal role in alleviating the metabolic burden on plants by restricting modifications to specific areas. The designed CRISPR-ESGE binary construct was introduced into Agrobacterium, which was then used to infect rice callus derived from seeds. The regenerated transformed plants were confirmed to contain the Cas12a gene, and the endosperm-specific expression of Cas12a was validated through RT-PCR analysis. Genome editing (GE) utilizing Cas12a offers distinct advantages over Cas9, which is characterized by heightened target specificity and decreased propensity for off-target effect, thereby potentially enhancing the grain starch content.

Abstract

The present invention relates to mesoporous amine-functionalized silica catalysts with worm-hole or channel-like morphology, prepared using 3-aminopropyl trimethoxysilane (3- APTMS) or 3-aminopropyltriethoxysilane (APTES). The catalysts exhibit average pore diameters of 1-100 nm and specific surface areas of 100-1,000 m2/g. The preparation process involves mixing Pluronic P123 with hydrochloric acid at 30-40°C, followed by addition of a mixture of 3-APTMS or 3-APTES with tetraethyl orthosilicate (TEOS), and subsequent hydrothermal treatment. The choice of precursor determines morphology: 3- APTMS yields worm-hole structures, while 3-APTES produces channel-like structures. These catalysts efficiently convert epoxides and carbon dioxide into cyclic organic carbonates with excellent yield. Additionally, the invention discloses a process for preparing styrene carbonate-coated zinc anodes and assembling full cells incorporating these anodes, significantly enhancing the durability of aqueous rechargeable zinc-metal batteries (ARZMBs).

Abstract

A novel class of phosphorus-containing Rucaparib analogues were synthesized and evaluated as poly(ADP-ribose) polymerase (PARP) inhibitors. Two distinct series were designed: one incorporating a spacer and the other without a spacer. Pharmacokinetic evaluation of representative compounds demonstrated favorable oral bioavailability and improved metabolic stability relative to Rucaparib. Oral administration of compound 9 (20 mg/kg, twice weekly) resulted in significant tumor growth inhibition (71%) compared to vehicle control and showed superior efficacy over Rucaparib (100 mg/kg, 52% inhibition). Importantly, compound 9 exhibited no mortality or significant body weight loss, while Rucaparib-treated animals displayed slight weight reduction and mortality in the final week of treatment. Collectively, these results highlight phosphorus-containing Rucaparib analogues as a promising new class of PARP inhibitors with favorable pharmacological properties and therapeutic potential for the treatment of BRCA-mutant and homologous recombination–deficient cancers, particularly breast and ovarian malignancies.

Abstract

The present invention relates to the method for efficient removal of phenolic compounds from wastewater generated from petroleum industry. The method is two-step process. The first step is a pre-treatment process which operates through adsorption-based separation using granular activated carbon. The wastewater in this step is allowed to pass through a column packed with granular activated carbon. In the second step, the pretreated wastewater is subjected to membrane-based treatment. The present invention also relates to the design of a special membrane for effective removal of phenolic compounds from waste water. The membrane formulation includes polysulfone as the main polymeric matrix and carbon dots coated powdered activated carbon as the additive which acts as the active agent for the removal of phenolic compounds. The membranes are designed in two different types, viz., (i) flat sheet and (ii) hollow fiber. The membrane-based separation using the flat sheet membranes can be done with the help of a dead-end filtration set up. In case of hollow fiber membranes, a cylindrical membrane module with a large number of hollow fiber membranes is designed. The membranes are reusable with excellent separation efficiency. The membrane module has proper feed inlet, outlet for permeates and outlet for retentates. This present invention provides removal of ~99% phenolic compounds from petroleum industry wastewater.

Abstract

Present invention discloses a novel green fluorescence silver nanodots prepared with naringenin, a flavanone, by a near green synthesis without adding harsh reducing agent such as sodium borohydride and others. Particularly, the present invention provides silver nanodots (quantum dot like nanostructure) of average size 3.5 nm. The preparation method is a unique kind of single pot green synthesis method wherein no common and strong reagent like sodium borohydride, ascorbic acid surfactant, thiol containing reagent or protein solutions are used. Alternatively, a simple flavanone, aromatic organic molecule that contains three OH groups is used both as a mild reducing and stabilizing agent for the preparation of the particles at room temperature (25°C). The aqueous suspension of the invented nanodots product is quite stable in room temperature. It produces strong green fluorescence with average fluorescence lifetime ~2.37 ns and produces a large stocks shift.
